Preparation and Characterization of Deoxycholic Acid-Conjugated Low Molecular Weight Water-Soluble Chitosan Nanoparticles for Hydrophobic Antimicrobial Peptide Carrier

Abstract
To develop the carrier of hydrophobic antimicrobial peptide, we introduced to hydrophobic group on low molecular water soluble chitosan (LMWSC). The deoxycholic acid (DA), is one the bile acid, was conjugated to LMWSC and then we prepared to nanoparticles using it. We confirmed the nanoparticle formation of deoxycholic acid conjugated LMWSC (LMDA) with dynamic light scattering and transmittance electron microscope. The particle size of LMDA was increased to decreasing the degree of DA substitution and its range was from 250 to 330 nm and had spherical shapes. We prepared to the loaded peptide, as an antimicrobial agent, LMDA nanoparticles (LMDA-A2). The release of antimicrobial agent from nanoparticles was slow and showed sustained release behaviors. Based on the results of release study that the higher DA contents in LMDA, the slower the releasing rate, particularly, the LMDA-A2 could be used as an excellent antimicrobial peptide carrier.
